The first published human trial of Melanotan II came out of a University of Arizona lab in 1996. Dorr and colleagues gave escalating subcutaneous doses to three male volunteers on alternating days over two weeks and reported that the peptide produced tanning activity after only five low doses (Dorr et al., Life Sciences, 1996, a pilot phase-1 study in three volunteers). The same paper noted side effects including nausea, drowsiness, fatigue and unplanned erections lasting one to five hours after injection, an early clue that the compound's effects reached beyond pigmentation.
That side effect became the subject of its own research line. A 1998 double-blind, placebo-controlled crossover study by Wessells and colleagues in the Journal of Urology gave Melanotan II to 10 men with psychogenic erectile dysfunction. Clinically apparent erections developed in 8 of the 10 men, well above the placebo response (Wessells et al., Journal of Urology, 1998, a double-blind placebo-controlled crossover study). Side effects in that trial included nausea, stretching, yawning and reduced appetite.
A larger 2000 study by the same lead author, published in the International Journal of Impotence Research, monitored 20 men with RigiScan equipment and found the peptide produced erections in 17 of them without any sexual stimulation. Reported sexual desire also increased more often after Melanotan II doses than after placebo doses in that trial (Wessells et al., International Journal of Impotence Research, 2000, a monitored dose-response study). Nausea was again the most common side effect, described as severe after a minority of standard doses.
A 2006 historical review by Hadley and Dorr in the journal Peptides traced this work as part of a broader research program into melanocortin peptides that began in the 1980s and 1990s, describing Melanotan II specifically as the compound that moved into clinical testing for erectile response (Hadley and Dorr, Peptides, 2006, a historical review). Later melanocortin research shifted toward more receptor-selective compounds, in part because Melanotan II activates several melanocortin receptor subtypes rather than just one, which the earlier trials link to some of its non-pigmentation effects.
None of the studies above describe how the compound was stored outside the clinical setting, so handling follows standard peptide practice rather than anything reported in the trials. Melanotan II is a lyophilised peptide and, like most peptides, sensitive to light and heat. The powder is kept cool and shielded from light before use, and once reconstituted with bacteriostatic water, the solution goes into the fridge and stays out of strong light between uses.
